The reduction of L-cystine in hydrochloric acid at mercury drop electrodes
نویسنده
چکیده
The reduction of L-cystine in 0.1 mol dm 3 HCl at 298 K has been studied at mercury electrodes. Dropping mercury electrode (DME), static mercury drop electrode (SMDE) and hanging mercury drop electrode (HMDE) modes were used with normal, sampled d.c. and differential pulse polarographic detection. The charge transfer kinetics for the irreversible reduction of L-cystine were complicated by reactant and product adsorption, by the formation of cysteinate complexes between mercury and the product thiol as well as by formation of Hg2Cl2. High Tafel slopes of 182 mV per decade were observed with a cathodic transfer coefficient of 0.32. The diffusion coefficient of L-cystine was found to be 5.3 · 10 10 m s 1 in 0.1 mol dm 3 HCl and 4.2 · 10 10 m s 1 in 2.0 mol dm 3 HCl at 298 K. The mechanism of L-cystine reduction at mercury has been discussed and the effects of pH and reactant concentration have been quantified. 2005 Elsevier B.V. All rights reserved.

The electroreduction of the disulphide bonds of insulin and other proteins.
Simple disulphides give cathodic waves at the dropping-mercury electrode which are due to their reduction to the corresponding thiols. The polarographic reduction of cystine has been studied by a number of authors (Brdicka, 1933b; Roncato, 1934; Kolthoff & Barnum, 1941; Kolthoff, Stricks & Tanaka, 1955). In acid solution it gives a single wave with a pronounced maximum, and in neutral or alkali...
متن کاملThe Optical Activity of Cysteine.*
The exact optical activity of cysteine has been a matter of doubt. Baumann (1) reported that [aID = -12.6” for the hydrochloride and -8.8” for the free ampholyte. Morner (2), on the other hand, stated that while some preparations of cysteine showed values similar to those of Baumann, other preparations were inactive or weakly dextrorotatory. Apparently no other investigator has reported dextror...

Voltammetric studies of rabbit liver metallothioneins (MTs) adsorbed onto thin mercury films preformed onto glassy carbon electrodes were carried out in MT-free phosphate buffers in an attempt to shed light on the possible redox-modulated MT metal-transfer processes. The redox behavior of the surface-confined MTs was studied by cyclic voltammetry and differential pulse voltammetry, and the amou...

متن کاملThe Determination of Cystine in Urine
At various times methods have been devised for the determination of cystine. Aside from the direct isolation of cystine, all the methods proposed for cystine entail the reduction of cystine to cysteine and determination of the latter. Most of the proposed methods are dependent on the reactive -SH group of the cysteine and accordingly give positive results with reducible disulfides other than cy...
